Interferon is a natural molecule that occurs in the body. It’s very important in modulating the immune system.  It has been shown to have a significant benefit in the treatment of Peyronie’s in multiple studies.

One of the advantages of Interferon injections is its safety profile.  Interferon can be used on any palpable plaque regardless of location.  

Interferon can be used safely for ventral curvature (typically caused by a plaque located close to the urethra) of the penis as well as for dorsal curvature

Important notice:  Interferon is no longer available in the US as of 2022, as its manufacturing was discontinued for business reasons (not enough sold). It may be available in some other countries.

How Effective is Interferon?

Interferon-a2B has been shown to have a good effect in the treatment of men with Peyronie’s disease in randomized controlled studies.  On average, men experience about 9% improvement;

  • overall 69% of men experience at least some improvement in curvature
  • 54% of men experience at least a 20% improvement in curvature (for the curvature of 40 degrees, 20% improvement is an 8-degree improvement, so curvature goes from 40 degrees to 32 degrees on average).

Research evidence shows that alpha interferon is effective in mild to moderate cases of Peyronie’s with a curvature up to 70 degrees. The side effects of the injection of interferon are uncommon and may present with flu-like symptoms in some people. Other less common side effects include hematoma, swelling, and edema.

How is Interferon Administered?

Interferon alpha-2B is administered by direct injection into Peyronie’s plaque similar to how verapamil and Xiaflex injections are performed.

The procedure is performed in the office under local anesthesia using a penile block, usually bupivacaine or lidocaine. After administration of local anesthetic, intralesional injections of interferon are nearly painless.  Local anesthesia also provides pain relief after injection. A compressive dressing wrap is placed on the penis for 12 hours.  Some men choose to get the injections without the administration of a local anesthetic.

The treatment schedule involves 6 biweekly injections, which may be repeated for up to 12 biweekly injections.

How Does Interferon Work in Treatment of Peyronie’s Disease?

Studies show that interferon decreases collagen production by fibroblast and stimulates collagenase activity (scar remodeling – essentially simulating the effect of Xiaflex).

What is the Treatment Course with Interferon?

  • Treatment is administered via direct injection into Peyronie’s plaque every 2 weeks.
  • 2 million U of interferon-a2b injected every 2 weeks for 6 to 12 treatments.
  • About 50% of men respond to therapy.
  • Absolute change of 9 degrees, a relative reduction of 20% from 42 degrees to around 30 degrees.
  • Treatment is effective for dorsal and ventral curvature.
